From my perspective (as a comic seller at cons) the top 4 characters are:
Batman
Deadpool
Harley Quinn
He walking dead dudes
So I'd guess I'd say deadpool or Harley Quinn. Lots of peeps looking for those books.

Miles Morales might end up as the most important character introduced since Wolverine, though I sincerely doubt it. I think he is also likely to be about as important of a character as Earth-2 Superman and Batman.
Storm, Colossus, and Nightcrawler are nearly as significant as Wolverine. Like it or not, so are Deadpool and Harley Quinn.
For that matter, so is James Rhodes. Due to James Rhodes appearance in three of the biggest blockbusters of the last five years, he is probably far more well known than Miles Morales.
Does this make these characters more important? I don't know. It depends how "important" is defined.
I have a hard time saying that Rick Grimes is an important chartacter. It is the title, not the character, that has significance. If there was a "early adventures of Rick Grimes" comic, it would probably not be an important book, or a great seller.
Teenage Mutant Ninja Turtles are in the running, I hadn't thought of them. I think Spawn is more significant than Rick Grimes is, that's for sure.
Let's also remember we live in a world in which Cerebus happened.
Sandman happened. How significant is Morpheus?

Gotta agree with Doc regarding Rick Grimes. Daryl i'd say is WAY more popular and he's not even in the comic. I do agree with Doc about Rick Grimes, that is why I listed the "walking dead dudes". Meaning the impact of TWD (the book) on the industry and popular culture is huge.
